Media differences exist in terms of callus fresh and dry weight, chromosomal behaviour and plant regeneration. It also appears that chromosomal behaviour of callus cells is assocoated with their regeneration capacity, particularly 0.1 mg/l kinetin exhibited minimum frequency of chromosomal aberrations with high frequency of plant regeneration. It suggests that cotyledon explant cultured on MS media supplemented with lower doses of phytohormones are more appropriate for Isolating plants with varying chromosome numbers aimed to induce somaclonal variation in vlgna spp.
