Present study was conducted on 100 lactating Murrah buffaloes maintained at LRC, NDRI Karnal (Haryana) to characterize and to identify genetic polymorphisms in HSPB1 gene. Two sets of primers specific to coding sequence of HSPB1 gene were designed using Primer3 software and PCR products of 631 bp and 670 bp were obtained. Amplicons were custom sequenced and subjected to ClustalW analysis which revealed 8 nucleotide changes, 7 in non coding region and one in coding region in Murrah buffalo sequence as compared to Bos taurus. Only one SNP at nucleotide position G225A in exon 1 of HSPB1 gene was observed in Murrah buffalo, which resulted in two genotypes GG and AG with respective frequency of 0.84 and 0.16 indicating the existence of variability in the sampled population. The study has opened the possibility of identifying and using genetic variations in major heat shock protein genes, for the future improvement of livestock through marker assisted selection.

Molecular characterization and SNP identification in HSPB1 gene in Murrah buffalo

<div>Background: The embryo genomic material used in this study came from an equal contribution of maternal and paternal genes. Gene expression during IVEP embryo development was controlled by several factors: the genetic makeup of the embryo, the epigenetic contributions and the culture materials. The aims of this research were to study the impact of ionomycin treatment on the sperm capacitation time and its impacts on the gene expression of some sensitive genes (Bcl-2, Bax, HSPB1, IGF1 and GDF9) during IVEP.&nbsp; 
 
Methods: The semen was placed in a sterile 15ml tube containing 5ml of capacitation medium and then split into three different experimental groups: the first group was the control; the second was the low-treated group, which contained sperm in a capacitation medium with 25 nM of ionomycin; the third group included 50 nM ionomycin.&nbsp; 
 
Result: The gene expression of BAX during early cleavage did not show any differences. However, in the late cleavage stage, the control group presented higher gene expression (P&lt;0.05) than that in the two treated groups. The Bcl-2 genes of the control group showed higher gene expression (P&lt;0.05) in both the early and late cleavage stages compared to the low-treated group. Gene expression of the HSP-1 gene was observed during early embryo development in the treated group (25 nM ionomycin). In conclusion, concentration 25 nM ionomycin showed higher gene expression in the early stages of embryo development also the best-quality embryos and gene expression results were found by studying gene expression. The transition from the maternal and paternal genome to the embryonic genome and gene expression during embryo development is controlled by several factors.&nbsp;</div>

Impact of Ionomycin on the Gene Expression of In vitro Fertilized Bovine Embryos

Background: Heat stress causes oxidative damage in chicken intestinal epithelial cells, resulting in economic losses in poultry production. The potential of resveratrol to alleviate oxidative stress and intestinal injury in poultry was investigated. 
 
Methods: A total of 300 one-day-old chickens were randomly assigned to five different experimental groups. The control group was maintained under normal temperature, while the remaining four groups were exposed to high-temperature conditions and received a basal diet supplemented with resveratrol for a period of 15 days. Intestinal tissue structure and serum antioxidant indices were measured. Mechanistically, transcriptome sequencing was employed to investigate gene expression changes in the jejunal mucosa. Finally, analyses of gene ontology (GO) and KEGG pathways were conducted. 
 
Result: The results indicate that heat stress negatively affects the structure of intestinal villi, leading to increased mRNA expression of HSPB1, HSP70 and HSP90. However, the daily administration of resveratrol at a dose of 400 mg/kg of body weight reduces the excessive expression of these heat shock protein mRNAs (P&lt;0.05). Chicks that were supplemented with resveratrol exhibited significantly elevated serum levels of SOD, CAT, T-AOC and GSH-PX (P&lt;0.05). Transcription profiling showed 181 significantly altered differentially expressed genes (DEGs). The outcomes indicated that the DEGs in the resveratrol-treated group were primarily enriched in the VEGF, ErbB and Toll-like receptor signaling pathways. All in all, resveratrol demonstrates significant potential in alleviating the adverse effects of heat stress on broilers.
